Deposition And Clearance Of A Water-Reactive Vapor, Methylphosphonic Difluoride (Difluoro), Inhaled By Rats

Abstract:
The deposition and clearance of inhaled methylphosphonic-difluoride (676-99-3) was studied in female F344-rats. Twelve to 16 week old animals were exposed by both inhalation and non inhalation routes to carbon-14 labeled methylphosphonic-difluoride or its hydrolysis products. Exposed animals were serially killed and dissected so that the movement of the radiolabel could be traced over 7 days. Constants were calculated for relating doses of the difluoride to various respiratory tract tissues as a function of total deposited vapor. Total deposition was estimated, and each organ was examined for amount of radioactivity. The deposition of inhaled difluoride was mainly in the maxilloturbinates and ethmoturbinates, and clearance was mainly by mucociliary clearance and swallowing. The constants indicated that the maxilloturbinates in the anterior of the nose received more than 4 times the dose received by the ethmoturbinates, while the trachea and lung received much lower doses than the nasal tissues. Hydrolysis products cleared rapidly after non inhalation administration. The authors conclude that the derived constants may be applicable for calculating doses to respiratory tract tissues for other inhaled vapors that do not saturate the mucus.

Abstract:
TD3: The teratogenicity of vaporized methlyphosphonic difluoride (DF) was investigated using the rat and rabbit as the experimental models. Pregnant rats and rabbits were exposed. Dams and does exposed exhibited nasal exudate and raspy breathing. These dams also had significantly lower body weights on the last three of four weighings. There were, however, no differences between the control litters and the DF-exposed litters for the parameters: litter number, mean body weight, mean crown-rump length, visceral or skeletal malformations. It was concluded that DF is not teratogenic in the rat or rabbit by the inhalation route at the concentrations tested. Keywords: Fetus; Teratogenic compounds; Gestation; Organogenesis; Pregnant. Abstract:
TD3: In 1987, the U.S. Army asked the National Research Council's Committee on Toxicology (COT) to review the literature that has been used as the basis to propose workplace exposure standards for four precursors of munition chemicals. The four precursors are methylphosphonic dichloride (DC), methylphosphonic difluoride (DF), pinacolyl alcohol (PA), and O-ethyl-O-(2-diisopropylaminoethyl)-methylphosphonite (QL). The Occupational Safety and Health Administration (OSHA) and American Conference of Governmental Industrial Hygienists (ACGIH) have not developed permissible exposure limits (PELs) or threshold limit values (TLVs) for these chemicals. The Army requested the review by COT to ensure that appropriate exposure standards are developed to protect workers and private citizens who might be incidentally exposed during chemical production, transportation, or storage. Abstract:
TD3: We present the 6-31G* optimized geometries and the corresponding vibrational frequencies of the three nerve agents sarin, soman, and tabun. Subsequently, we correct the computed frequencies of the three molecules by multiplying the computed frequencies by previously derived correction factors. We compute the 6-31G* optimized geometries and vibrational frequencies of methylphosphonic difluoride and methylphosphonofluoridic acid to derive correction factors for the P-F stretching and bending vibrational frequencies. Abstract:
TD3: The development of a procedure for the recording of reference mass spectra of trimethylsilyl (TMS) esters of alkyl and cycloalkyl methylphosphonates is described. The compounds are prepared in situ, on a small scale, from methylphosphonic difluoride and a selected alcohol dissolved in acetonitrile. Hydrolysis of the formed alkyl and cycloalkylmethylpho sphonofluoridates to the corresponding methylphosphonates, evaporation to dryness and trimethylsilylation of the residue produces the TMS esters. The resultant reaction mixture is analysed by gas chromatography-mass spectrometry. In this way, electron impact mass spectra of TMS esters of 17 alkyl (ranging from butyl to nonyl) methylphosphonates and five cycloalkyl methylphosphonates were recorded.The mass spectra and a short description of the fragmentation processes of the TMS esters are presented.
