2000 - TOXICITY OF REFRIGERANTS (SANITIZED) Acute inhalation toxicity was evaluated in guinea pigs (sex, strain, and number not reported) receiving chlorodifluoromethane at concentration levels of 4.4% (v/v) for 6 hours and 5.2 or 20.3% (v/v) for 2 hours. No mortalities were reported. Slight labored breathing, and shivering were noted in 1 animal of the 4.4% group. At 5.2% there were occasional chewing movements, partially closed eyes, irregular breathing, shaking of heads, and rubbing of noses with feet. All animals at this exposure level recovered (time of recovery not reported). At 20.3% animals showed inability to stand, convulsive leg movements, tremors, lacrimation,
nasal discharge, and audible breathing. All recovered within 1 day of exposure. Gross necropsy findings were not reported.

2000 - ARCTON 22: SECOND DOMINANT LETHAL STUDY IN THE MOUSE WITH ATTACHMENTS

The dominant lethal effects of chlorodifluoromethane were evaluated in 6 groups of 20 fertile male CD-1 mice exposed by inhalation to chlorodifluoromethane (Arcton 22) at nominal concentrations of 10, 100, 500, 1000, 10000, or 50000 ppm for 6 hours/day, for 5 consecutive days. A group of 40 male mice served as the untreated control. A group of 20 mice injected once intraperitoneally with 200 mg/kg cyclophosphamide served as the positive control. Mating followed the final treatment period. Each male mouse in each treatment group was paired with 2 virgin female 10-week old mice, during 8 consecutive 4-day mating periods, commencing at 7-day intervals. A significant reduction in the percentage of females which became pregnant occurred in the 5th mating period in females mated to males in the 10,000 and 50,000 ppm exposure groups. The number of successful matings per male was statistically significantly reduced in the 10 ppm exposure group in weeks 4 and 7, in the 10,000 ppm group in week 5, and in the 50,000 ppm group in weeks 1, 5, and 7. The time of sacrifice of the mated females was not reported. The number of implantations per pregnancy was significantly reduced in females mated to males in the 100 ppm exposure group (week 4), in the 500 ppm group (week 6), and in the 1000 ppm exposure group (week 5). All treated groups except the 10,000 ppm group had a significant increase in early deaths. Late deaths were significantly increased in pups of females mated in week 4 (100, 1000, and 50000 ppm) and in week 7 (50000 ppm).

2000 - ARCTON 22: A SECOND CYTOGENETIC STUDY IN THE RAT WITH ATTACHMENTS

The frequency of chromosome aberrations was determined in bone marrow cells of Wistar-derived male rats exposed by inhalation to chlorodifluoromethane (Arcton 22) for 6 hours/day over 5 consecutive days. Five groups of 12 male rats (150 to 200g; 8 to 10 weeks old) were exposed to nominal concentrations of 0, 10, 100, 500, and 1000 ppm of Arcton 22. A positive control group of 12 rats was exposed to 4000 ppm benzene. Cell division was arrested 4 hours after the final exposure with an intraperitoneal injection of colchicine (3 mg/kg). Rats were sacrificed 6 hours after the final exposure, and fifty cells from each animal were examined. The proportion of cells with any abnormality, and the proportion of cells with any abnormality except gaps, were compared with the negative control group using ANOVA (data were arcsine-transformed). The proportion of animals with any abnormality except gaps was compared with the negative control using Fisher's exact test. There was an inverse dose-response relationship between treated groups. The mean percentage of bone marrow cells with chromosomal abnormalities (other than gaps) was significantly different in the group nominally exposed to 10 ppm, but not at higher concentrations. The percentage of rats with chromosomal abnormalities (except gaps) was elevated in groups exposed to all but the highest concentration.

Chlorodifluoromethane (FC-22) was evaluated for embyotoxicity and teratogenicity in groups of 40 pregnant Charles River rats exposed to the test substance by inhalation at concentrations of 0, 0.05, 0.10, and 2.00% on days 6-15 of gestation. No clinical signs of toxicity were observed in maternal animals. The number of implantations, early and late resorptions, and number of live fetuses per litter were unaffected. There was a sporadic appearance of major malformations of the eye in all test groups. The increased incidence of eye defects was not statistically significant. Authors believe that the test substance may have interacted with the genetic make-up of affected fetuses and caused the increased expressivity of a mutant gene. The authors considered the test substance to be a mutagen under the conditions of this study.

1992 - Fatal Accident Circumstances and Epidemiology (FACE) Report: Assistant Manager at Ice Rink Asphyxiated by an Oxygen-Deficient Atmosphere, Alaska, May 20, 1991.

The case of a 24 year old male assistant manager for a shopping mall ice skating rink who was asphyxiated inside a compressor room while attempting to shut off a refrigerant gas leak was examined. The gas was chlorodifluoromethane-22 (75-45-6) (CFC-22). The employer was the owner of a 170 store indoor shopping mall which included a swimming pool and an ice skating rink. There was no safety policy or program in place. The refrigeration system had a long history of leaks. A large leak had been plugged in a makeshift fashion. A maintenance worker performing a routine check observed refrigerant oil oozing from under the doors to the compressor room. The victim, the maintenance supervisor, and a maintenance worker entered the compressor room through self closing doors. The victim was wearing a cartridge type respirator which was inadequate in an oxygen deficient atmosphere. A coworker called 911 after entering the compressor room and seeing two of the workers lying on the floor. The maintenance worker [abstract truncated]

1990 - A STUDY OF THE RATE OF UPTAKE AND ELIMINATION OF CHLORODIFLUOROMETHANE (HFA22) IN MAN BY THE INHALATION ROUTE WITH COVER LETTER DATED 02-07-90

ICI CENTRAL TOXICOL LAB

HFA22 (chlorodifluoromethane) was evaluated for pharmacokinetics in groups of 3 male humans exposed via inhalation to vapor concentrations of 92 or 518 ppm for 4 hours in an inhalation chamber. Blood and expired air samples were taken during exposure and for up to 26 hours after exposure. Urine samples were taken for up to 22 hours after exposure. The compound was absorbed very quickly, reaching plateaus of 0.25 and 1.36 ug/ml after approximately 4 and 3 hours of exposure for the low- and high-dose levels, respectively. End-tidal breath concentrations of the test compound during exposure were similar to the exposure concentration
s, indicating that only a small proportion of the available dose was absorbed. Elimination of the compound from blood was triphasic, with estimated half-lives of 0.005, 0.20, and 2.7 hours. The first phase probably corresponded to elimination from blood and rapidly perfused tissues, whereas the second and third phases of elimination probably corresponded to elimination from slowly perfused tissues and fat, respectively. The parent compound was detected in urine samples from all subjects; very little fluoride ion was detected in urine samples, indicating that the test compound was probably not metabolized to a significant extent.

Arcton 22 (chlorodifluoromethane) was evaluated for clastogenicity in C57BL/6JfCD-1/Alpk mice exposed to vapor concentrations of 0 (5 mice/sex), 50,000 (5 mice/sex) or 150,000 ppm (15 mice/sex) for 6 hours in a dynamic air-flow chamber. Mice were sacrificed and bone marrow was examined for micronucleated polychromatic erythrocytes at 24 hours after treatment with 50,000 ppm, and 24, 48, or 72 hours after treatment with 150,000 ppm. The frequency of micronucleated polychromatic erythrocytes did not increase in any of the treatment groups, indicating that Arcton 22 was not clastogenic in mice under the conditions of this assay.

Teratogenicity was evaluated in 4 groups of 19 pregnant CD female rats receiving Arcton 22 via inhalation at concentration levels of 0, 100, 1,000 and 50,000 ppm for 6 hours per day on gestation days 6 through 15. There were no treatment-related effects in appearance, behavior, mortality, or pregnancy rate. At 50,000 ppm maternal weight gain was slightly lower than the control. There were no effects on body weight at 100 or 1,000 ppm. In all test groups litter size, post-implantation loss, litter wight, and mean fetal weight were similar to the control. At 50,000, there was an increased incidence of anophthalmic fetuses. Keywords:

1989 - EFFECT OF ARCTON 22 ON PREGNANCY OF THE NEW ZEALAND WHITE RABBIT WITH ATTACHMENTS AND COVER LETTER DATED 07-05-89
Teratogenicity was evaluated in 4 groups of 14 to 16 pregnant New Zealand white female rabbits receiving Arcton 22 (chlorodifluoromethane) via inhalation at concentration levels of 0, 100, 1,000 and 50,000 ppm for 6 hours per day on gestation days 6 through 18. There were no signs of treatment-related effects on mortality or pregnancy rate. At the highest concentration level there was slight weight loss during the first 4 days of exposure, but weight gains were comparable to controls. Litter values (determined by litter size, implantation loss, litter and mean fetal weights) were similar to the control at all exposure levels.
There were no signs of treatment-related embryonic or fetal malformations noted. Parameters such as the number of corpora lutea, implantations per dam, resorption rates, fetal weights and or sex distribution were not reported

The ability of chlorodifluoromethane to cause mutation at the gene locus coding for hypoxanthine-guanine phosphoribosyl transferase in Chinese hamster ovary cells (CHO/HGPRT Assay) was evaluated in the presence and absence of metabolic activation from Aroclor-induced rat liver S9 fraction. Results of preliminary toxicity tests of non-activated cultures treated with concentrations of 0, 20, 31, 33, 58, 60, 73, 75, 83, and 92% chlorodifluoromethane produced plating efficiences ranging from 115.9 to 93.2% relative to the nitrogen gas control. Assays of activated cultures at concentrations of 0, 32, 36, 60, 70, 76, 78, 81, and 82% produced cloning efficiencies ranging from 109.9 to 78.3% relative to the nitrogen gas control. The mutagenicity of chlorodifluoromethane was evaluated in Salmonella tester strains TA98, TA100, TA1535, and TA1537 (Ames Test), both in the presence and absence of added metabolic activation by Aroclor-induced rat liver S9 fraction. Based on the results of preliminary bacterial toxicity testing, the material was tested for mutagenicity at concentrations of 0, 15, 25, and 40% (of the atmosphere) using the dessicator method. The material caused a reproducible positive response in tester strain TA1535, both in the presence and absence of S9 activation. Chlorodifluoromethane did not cause a reproducible positive response in tester strains TA98, TA100, and TA1537, either in the presence or absence of added metabolic activation.

Based on the results of preliminary bacterial toxicity testing, the material was tested for mutagenicity at concentrations of 0, 20, and 40% (of the atmosphere) using the dessicator method. Chlorodifluoromethane did not cause a reproducible positive response in any of the bacterial tester strains, either in the presence or absence of added metabolic activation.

The mutagenicity of 1,1,1-trichloroethane (TCE) was evaluated in Salmonella tester strain TA100 (Ames Test, desiccator assay variation), both in the presence and absence of added metabolic activation by Aroclor-induced rat liver S9 fraction. TCE was tested for mutagenicity at dose levels of 0.4, 1.90 and 8.92 gm/desiccator. No rationale for selection of dose levels used was reported. TCE caused a positive response in strain TA100 with metabolic activation at 1.90 ml/desiccator. The mutagenicity of T-1716AC (60% TCE, 30% monochlorodifluoromethane, and <2% 1,4-dioxane) was evaluated in the same strain under the same conditions as abov e. Although the dose levels were not reported, the authors did state that the mixture was discharged for time periods of 0.5, 1.0 and 5.0 seconds. T-1716AC did not cause a reproducible positive response in TA100 at any of the exposure periods.

1980 - ACQUISITION AND CHEMICAL ANALYSIS OF MOTHER'S MILK FOR SELECTED TOXIC SUBSTANCES (DECEMBER 1980)

Samples of mother's milk were collected from persons in 5 cities and analyzed by gas chromatography and spectrophotometry for volatile and semivolatile organics. In the volatile fraction, 26 halogenated hydrocarbons, 17 aldehydes, 20 ketones, 11 alcohols, 2 acids, 3 ethers, 1 epoxide, 14 furans, 26 other oxygenated compounds, 4 sulfur-containing compounds, 7 nitrogen-containing compounds, 13 alkanes, 12 alkenes, 7 alkynes, 11 cyclic hydrocarbons, and 15 aromatics were found, including major peaks for hexanol, limonene, dichlorobenzene, and some esters. Semivolatile compounds of interest that were identified in the samples were polychlorinated biphenyls and DDE. Polychlorinated naphthalenes, polybrominated biphenyls, chlorinated phenols, and other compounds were specifically sought but not detected (limit of detection was about 20-100 ng/mL mild).
